a) Introduction: Cannabis sativa is composed of more than one hundred compounds, including terpenes, phenolic compounds, and cannabinoids, like Δ9-THC and CBD, which are extracted in alcoholic extracts, which we refer to as Cannabis Alcoholic Extracts (CAE). One strategy for administering CAE is chitosan nanocapsules (NCs), which have an oily core and a hydrophilic shell. One strategy for administering CAE is chitosan nanocapsules (NCs), which have an oily core and a hydrophilic shell. These offer biocompatibility, biodegradability, and mucoadhesiveness. b) Objective: This study aims to optimize the synthesis of a CAE-carrying nanocarrier, characterize its morphology, and determine it´s encapsulation efficiency (EE). c) Methodology: The synthesis was carried out using the solvent displacement method. The organic phase comprises olive oil, Tween80, and acetone. Conversely, the aqueous phase is a chitosan solution. The parameters were optimized using a factorial design through Minitab18 software. Particle size, PDI, and Z-potential were determined using DLS, and the morphology was analyzed by SEM, where the samples were coated with a layer of Au/Pd before analysis. EE was calculated by quantifying CBD by HPLC. d) Results: The synthesis parameters were effectively improved, leading to NCs with a size of 231 nm, a PDI of 0.18, and a Z-potential of +34 mV. SEM analysis showed that the NCs had a spherical morphology. The EE was determined to be 98%. e) Conclusion: The results showed that the synthesis of the proposed nanosystem was effectively achieved, with suitable nanometric parameters and high EE.
